Referenzmaterialien für die Pharmakogenetik (PGx)
SensID hat eine vollständig plattformunabhängige (agnostische) Kontrolle für Dihydropyrimidin-Dehydrogenase-Tests (DPYD; DHP; DPD; DHPDHASE) entwickelt. Die enthaltenen Varianten wurden auf der Grundlage von Publikationen und Vorschlägen verschiedener Arbeitsgruppen und Experten ausgewählt, die die Implementierung des DPYD-Tests unterstützen. Behörden wie EMA und NHS empfehlen, alle Patienten vor einer systemischen Therapie mit den FU-haltigen Medikamenten 5-Fluorouracil (5-FU), Capecitabin, Tegafur und Flucytosin auf eine DPYD-Mutation zu testen. Dieses Produkt besteht aus 3 separaten Artikeln gDNA mit entweder 0%, 50% oder 100% mutiertem Gen mit einem besonders gut charakterisierten genomischen Hintergrund. Mutationen in diesem Gen führen zu einem Defekt der DPYD, und damit zu einem Fehler im Pyrimidinmetabolismus, der mit Thymin-Uracilurie assoziiert ist, wodurch ein erhöhtes Toxizitätsrisiko bei Krebspatienten besteht, die eine 5-Fluorouracil-Chemotherapie erhalten.
Therapeutische Indikationen, bei denen der DPYD-Mutationsstatus relevant ist:
- kolorektales Karzinom
- Magentumore, Bauchspeicheldrüsenkrebs, Speiseröhrenkrebs
- Brustkrebs
- Plattenepithelkarzinom des Kopfes und Halses
- Gallenwegskarzinome
- Nicht-kleinzelliger Lungenkarzinome (NSCLC)
- Systemische Hefe- und Pilzinfektionen
Das Produkt ist ideal geeignet für Next Generation Sequencing (NGS) Arbeitsabläufe. Insbesondere zur
– Validierung und Entwicklung von Sequenzierprotokollen (z.B. Bereich Whole Exome Sequencing (WES), Amplicon Sequencing) und PCR-Protokollen
– Kalibrierung und Entwicklung von Geräten und Arbeitsabläufen zur DNA Aufbereitung (z.B. DNA Fragmentierung mittels Schall oder enzymatischem Verdau)
– Vergleich der eigenen NGS Performance mit frei verfügbaren Datensätzen
